Easier and Harder have regular gameplay. The other (Silly) contains some unusual elements, and should not really be taken seriously. Your main objective is to obtain loot. There is also a "return home" objective. On "Easier", there is an optional no kill objective, and on "Harder", there is an enforced no kill/3 knockout limit objective. The limit on knockouts was added because the mission is still too easy without it. There are a couple of additional objectives on "Silly", but you can see those for yourself in-game.
